It depends on what job you are looking for?

if you look for working as helpdesk, you need to know windows troubleshooting, solving email issues in outlook, solving internet connectivity, also troubleshoot applications check the processes needed to be running for certain application, also its important to have customer services skills which reflect your company's SLA. also working with helpdesk softwares.you need Comptia A+ certificate to start such job.

If you applying for IT technical support, you need to know hardware and software troubleshooting, installing and configuring laptops, desktops, AP's, CCTV, IP cams, cabling, troubleshooting some switches issues like changing the VLAN for a computer, schedule updates for antivirus system, you have to understand the needs and softwares and configurations for each certain computer used by each employee depending on his job.you need to setup and create, edit user accounts on AD and exchange or office 365.

you need A+ and CCNA and MCSA  in addition to backup solutions to start this job .

if you applying for systems engineer you need to excel systems like windows, linux or unix, solari, setup, configuring and troubleshooting in addition to VMware and cloud solutions....

to start this job you need CCNA data center, MCSA, RHCSA, Exchange, VMware certifications,...

if you applying for programming jobs:

web development: HTML, CSS, HTML 5, JQuery, Ajax, XML, ASP.net or php,...

Mobile development: Android development using java, HTML5 ,.... C to develop Apple applications,...
