What is the partition chromatography?

- So this chromatographic technique is the first kind of HPLC ever used.

 

- It uses the solubility of the analytes (molecules to be separated) in two immiscible (non-mixing) liquids. One of the liquids is used as the eluant, the other is used as the stationary phase.

 

- The analytes partition, or move back and forth, between the eluant and the stationary phase at certain rates, depending on the characteristics of each analyte.

 

- Therefore, certain analytes will elute first because they solvate in the eluant at a faster rate, while other analytes will elute last, due to their higher solubility in the stationary phase.

Partition Chromatography mean by " solute mixing with mobile phase and moving through the stationary phase at the time the solute partition between mobile phase and stationary phase. It is called Normal Phase chromatography.

 

This form of chromatography is based on a thin film formed on the surface of a solid support by a liquid stationary phase.
Solute equilibrates between the mobile phase and the stationary liquid.
